Not necessarily. That applies to anyone with a Chaotic Evil alignment, like an orc or something, but a redshirt orc clearly couldn't be considered a supervillain. Similarly, a supervillain might well have a single clear goal (taking over the world being the classic). There are two parts of a supervillain: the super and the villain. A supervillain has to have, if not superpowers or giant robots or stuff, then at least the intelligence and resources to actually be a threat on a large scale. A supervillain has to be thoroughly evil, as opposed to an antihero or someone of a Neutral alignment, who might do bad things but isn't necessarily bad to the core.
